Semen biomarker TEX101 predicts sperm retrieval success for men with testicular failure

Background: Azoospermia could be due to either obstruction (obstructive azoospermia: OA) or spermatogenic failure (non-obstructive NOA). Close 50% of men with NOA have small pockets sperm in the testis which retrieved surgically and then injected into oocytes a program intra-cytoplasmic insertion. Presently, there are no accepted non-invasive tests allowing clinicians predict success rates retrieval. Previously, we identified germ cell-specific protein TEX101 semen found primary spermatocytes more mature forms, but not spermatogonia, Sertoli Leydig cells. We hypothesized that concentration used production NOA. Methods: This was prospective cohort study on being treated at male infertility centre. Men planning retrieval provided 1–3 samples prior surgery. Semen concentrations were measured by an in-house-developed ELISA assay compared results surgery retrieve sperm. Results: 20/60 karyotypically normal had < LOD (<0.2ng/mL). Of these, 0% successful retrieval(0-17%: 95% CI) . In contrast, 40 TEX101> LOD, (34-66%: CI, sig diff. Fisher’s exact test, p<0.05). Conclusions: Undetectable (<0.2 ng/mL) is highly predictive for single strongest predictor reported so far. will help couples decide their individual chances retrieval.
